| Background | DOG1 (protein kinase C theta), the novel marker, is expressed ubiquitously in gastrointestinal stromal tumors irrespective of kit or PDGFR alpha mutation status. Reactivity for DOG1 may aid in the diagnosis of GISTs, including PDGFRA mutants that fail to express KIT antigen, and lead to appropriate treatment with imatinib mesylate, an inhibitor of the KIT tyrosine kinase. |

| Applications | Formalin-fixed paraffin-embedded as well as acetone fixed cryostat tissue sections. No further titration is required. We suggest an incubation period of 30-60 minutes at room temperature. However, depending upon the fixation conditions and the staining system employed, optimal incubation should be determined by the user. Formalin fixed paraffin embedded tissue sections require high temperature antigen unmasking with 10 mM citrate buffer, pH 6.0 prior to immunostaining.
