| Background | D4-GDI belongs to the family of Rho-GDIs or GDP-dissociation inhibitors for the Rho / Rac family of small G proteins; consisting of RhoGDI-1, D4-GDI / RhoGDI-2 and RhoGDI-3. It inhibits the release of GDP from the Rho proteins, essential for GTP-loading and activation of GTPase activity. D4-GDI is located in the cytoplasm and is involved in the regulation of the membrane asoociation and dissociation cycle of Rho / Rac proteins. Rho family of G proteins is involved in reorganization of actin cytoskeleton. Rho-GDIs are expected to play an important role in its spatial determination. D4-GDI is known to be a substrate for Caspase 3 during Fas-mediated apoptosis. Its cleavage may contribute towards the cytoskeletal changes involved during apoptosis. |

| Applications | Western Blotting: Ab 5-10 µg/ml for 2 hrs at RT.
Immunohistology (Formalin/paraffin): Use Ab at 5-10 µg/ml for 30 min at RT. Staining of formalin-fixed tissues requires boiling tissue sections in 10mM citrate buffer, pH 6.0, for 10-20 min followed by cooling at RT for 20 min. Recommended positive control: Jurkat Cells, Placenta. |
