| Background | Major vault protein (MVP) is identical to lung-resistance related protein (LRP). Its shape is reminiscent of the nucleopore central plug. Treatment of cells with estradiol increases the amount of MVP in nuclear extract. The hormone-dependent interaction of vaults with ER is prevented in vitro by sodium molybdate. Antibodies to progesterone and glucocorticoid receptors are also able to co-immunoprecipitate the MVP. Reportedly, the lung-resistance related protein (LRP) is identical to human MVP. LRP is overexpressed in many neoplastic tissues and cell lines indicating a poor response to chemotherapy. MVP is part of the ribonucleoprotein complex, which is highly conserved in all eukaryotic cells. |

| Applications | Immunohistochemistry on paraffin embedded sections (1:200 - 1:400). Staining of formalin-fixed tissues REQUIRES boiling tissue sections in 10mM citrate buffer, pH 6.0, for 10-20 min followed by cooling at RT for 20 min. Positive Control: SKBR-3 cells. Breast tumors.
Western Blot (1-2µg/ml for 2hrs at RT). Immunoprecipitation (Native): Use Protein G and Ab at 2 µg/mg protein lysate. Flow Cytometry and Electrophoretic mobility shift assays (EMSA). |
| Specificity | Reacts with a 104 kD MVP, which is known to interact with the estrogen receptor. Species: Human and Rat. |
