| Background | DFF45 is the human homologue of mouse ICAD which was identified as an inhibitor of caspase-activated deoxyribonuclease (CAD). ICAD/DFF45 is a downstream regulator of DNA degradation. CAD is an apoptotic DNase with high specific activity and exists as an inactive complex with ICAD/DFF45 in living cells. Upon apoptosis, caspase 3 is activated and cleaves ICAD/DFF45, which releases CAD from ICAD/CFF45. CAD then could enter the nucleus to degrade the chromosomal DNA.
ICAD/DFF45 may inhibit not only the DNase activity of CAD, but also its translocation into the nucleus. Since functional CAD can only be synthesized in the presence of ICAD/DFF45 in a cell-free system, ICAD/DFF45 seems to function as a chaperone for CAD during its synthesis. |

| Specificity | The 6B8 monoclonal antibody reacts with human ICAD/DFF45 n Western blot.
Using total cell lysates from Jurkat, Raji or U937 cells, this antibody detects two bands of 45 kDa and 40 kDa corresponding to human ICAD-L and ICAD-S respectively. The 6B8 antibody does not crossreact with mouse ICAD. |
| Storage | Store lyophilized (preferably in a desiccator) at 2-8°C and reconstituted (in aliquots) at -20°C.
