| Applications | Immunofluorescence: 1/400 - 1/800 for IF microscopy using FITC with overnight incubation at 2-8°C.
Immunohistochemistry on frozen sections and paraffin embedded tissues (proteolytic treatment recommended - see protocol). Recommended positive control: Gut (peripheral nerves), fetal lung. Working dilution: 1:50 for immunohistochemistry, dilute immediately before use. Dilution buffer: PBS with 1% albumin and 0.1% Na-Azide. Incubation time: 30 min at room temperature or overnight at 2-8°C. |

| Protocols | Protocol for the use of anti-Bombesin polyclonal antibody on paraffin-embedded tissue sections
FIXATION: Small tissue blocks are fixed in 4% buffered formaldehyde (pH 7.6) at 4°C. The fixation time should not exceed 12 h. During the embedding process temperatures should not exceed 58°C. STAINING PROCEDURE: Deparaffinize and rehydrate sections. If necessary, block endogeneous peroxidase activity (0.3% H2O2 in 50% methanol, 15 min). Rinse in PBS 3x. Incubate for 5 min in 0.1% pronase in PBS at 37°C. Rinse in PBS 3x. Apply properly diluted anti-Bombesin antibody (1:50) and incubate for 30 min at room temperature or overnight at 4°C. Rinse 3x. Apply conjugated antibody. Rinse in PBS 3x. Apply substrate. |
