| Background | Laminins are glycoproteins of the extracellular matrix, especially of the basal membranes. They are multi-functional molecules which play an important role during development and differentiation of organs and for cell movements. These properties rely on the interaction of laminins with receptors on cell surfaces like integrins, collagen IV, proteoglycans. Laminins consist of 2 heavy chains (A and M, 300-400 kDa) and 1 light chain (B1, B2 or S, 200 - 220 kDa). |

| Applications | IF: 1:40.
IHC: 1:200 - 1:600 for paraffin sections, 60 min at RT or 2-8°C over night. ELISA. RIA. Positive control: mouse skin. Pretreatment: After de-waxing the tissue slices they are treated with 0.2% hyaluronidase (app. 300 U/mg) in TBS 15 min at 37°C. There after non-specific binding is blocked by blocking serum or 3% BSA in TBS. For peroxidase systems blocking with 1% peroxide solution in TBS for 30 min at RT is recommended. |
| Specificity | Murine laminin 100%, murine collagen type I, II, III, IV < 0.1 %, murine fibronectin < 0.1 % (RIA at 1:40000 dilution). The antibody against laminin primarily labels basal membranes (especially s. c. lamina lucida) and may be used to detect disturbances in the basal membrane during tumour invasion. Laminin is also found in extracellular matrix not related to basal membranes, e.g. in the proximity of neurons especially during development, regeneration and tumour development. |
| Storage | Store the antibody at 2 - 8 °C up to one month or (in aliquots) at -20 °C for longer. Avoid repeated freezing and thawing.
