| Background | c-Abl is a nonreceptor tyrosine kinase that is located in either the nucleus or the cytplasm depending on the splice isoform (1a and 1b). c-Abl is implicated in cell differentiation, division, adhesion and stress response (2). C-Abl is composed of a catalytic domain, a SH2 and SH3 domain and an actin binding domain. Alteration of ABL by chromosomal rearrangement (BCR-ABL) or viral transduction (v-Abl) causes chronic myeloid leukemia (CML) and leads to malignancy (3). cAbl has been known to play a role in the insulin signaling pathway through its interaction with SORBS1. It has also been shown that that the tyrosine kinase activity of nuclear c-Abl is regulated in the cell cycle through a specific interaction with the retinoblastoma protein (RB1) (4); and can be activated by ionizing radiation and other DNA damaging agents such as DNA-PK and ATM (5). |
