| Application | Western Blotting: 5-10 µg/ml for 2 hrs at RT. Immunohistology (Formalin/Paraffin): Use 1/25-1/50 for 20 min at RT. Requires boiling tissue sections in 10 mM citrate buffer, pH 6.0, for 10-20 min followed by cooling at RT for 20 min. Recommended Positive Control: Conditioned, serum-free medium from (TPA-treated) HFL-1 cells. Placenta, Bladder, Breast, Ovarian Carcinoma. |
| Background | Matrix metalloproteinases (MMP) are proteolytic enzymes capable of degrading connective tissue components. Imbalanced secretion of certain MMP or disturbances in the differential control of MMP by tissue inhibitor of MMPs (TIMPs) are implicated in the invasive potential of malignant tumors. Interstitial collagenase (MMP-1), 72 kDa gelatinase A/type IV collagenase (MMP-2), and 92 kDa gelatinase B/type IV collagenase (MMP-9) dissolve extracellular matrix (ECM) and may initiate and promote angiogenesis. |
