| Application | Immunohistochemistry on Paraffin Sections: 1/100 for 30 min at RT. Staining of formalin-fixed tissues requires boiling tissue sections in 10mM citrate buffer, pH 6.0 for 10 min followed by cooling at RT for 20 min. Slides must be washed in between steps. Rinse slides with PBS/0.05% Tween. Positive Control: Placenta, Bladder, Breast, and Ovarian carcinoma. |
| Background | MMPs are proteolytic enzymes capable of degrading connective tissue components. They have a common mode of activation, a conserved amino acid sequence in the putative metal binding-active site region, and are inhibited by specific tissue inhibitors of metalloproteinases (TIMPs). MMPa and TIMPs play a significant role in regulating angiogenesis. MMP-2 is synthesized as a 631 amino acid proenzyme which is activated by cleavage of the first 80 amino acids. |
