Application	Western Blotting: 1-2 µg/ml for 2 hrs at RT. Immunohistology (Formalin/paraffin): 1-2 µg/ml for 30 minute at RT, staining of formalin-fixed tissues is IMPROVED by boiling tissue sections in 10 mM citrate buffer, pH 6.0, for 10-20 min followed by cooling at RT for 20 min. Positive Control: Conditioned, serum-free medium from (TPA-treated) human fetal lung cell line HFL-1. Bladder, breast, and ovarian carcinoma.
Background	Tissue inhibitor of metalloproteinases-1 (TIMP-1) and Tissue inhibitor of metalloproteinases-2 (TIMP-2) are inhibitory enzymes of matrix metalloproteinase family, and are thought to be of great importance in the maintenance of connective tissue integrity. TIMP-1 forms a complex of 1:1 stoichiometry with activated interstitial collagenase, activated stromelysin, active form of 72kDa Type IV collagenase (also known as MMP-2 or gelatinase A), and latent and active forms 92 kDa Type IV collagenase (also known as MMP-9 or gelatinase B). TIMPs inhibit the proteolytic invasiveness of tumor cells and normal placental trophoblast cells.
