| Background | TIMP-1 is expressed by a wide range of cells including microvascular endothelial cells, smooth muscle cells, mesangial cells and some fibroblasts. TIMP-1 inhibits matrix metalloproteinase activity by forming complexes via reversible non covalent binding. It is reported to be involved in the control of angiogenesis and tumour growth. |

| Protocols | Staining of Paraffin Embedded Sections: For normal soft tissues, bake slides for one hour at 60°C. Bone requires longer baking time (overnight). Deparaffinize tissues in xylene, and rehydrate through graded alcohols. Rinse with distilled water. Quench endogenous peroxidase in 3% hydrogen peroxide for 10 minutes. Rinse in PBS Microwave in a 0.01 M citrate buffer. Rinse in PBS and digest in 0.1% trypsin at 37°C for 20 minutes. Rinse again in PBS. Block non-specific binding with 1/20 sheep serum. Rinse with PBS Drain slides and incubate overnight at 4°C with appropriate dilution of BP858. Rinse with PBS Incubate with appropriate dilution of sheep anti rabbit secondary antibody. Rinse with PBS Incubate with streptavidin HRP Rinse with PBS Detect colour reaction with AEC chromogen for 5 minutes Rinse with distilled water Counterstain with Meyers Hematoxylin Rinse with distilled water Blue with ammonia water Rinse with distilled water Coverslip with aqueous montin |
