Antibodies to DNA Fragmentation Factor: DFF40 (CAD) and DFF45 (ICAD)

Introduction

The DNA fragmentation factor (DFF) is composed of two subunits, the 40-kDa caspase-3-activated nuclease (DFF40/CAD) and its 45-kDa inhibitor (DFF45/ICAD).

Apoptosis is induced through a number of cell death receptors and their ligands like TNF and TNF receptor. The signaling cascade is carried further by adaptor proteins containing death domains and by caspase family of proteins. The caspase-activated DNase (CAD) is the primary nuclease responsible for oligonucleosomal DNA fragmentation during apoptosis. DFF45/ICAD acts as a chaperon for appropriate folding of DFF40 and it also acts as inhibitor of DFF40. Caspase 3 upon activation cleaves DFF to release DFF40/CAD, which oligomerizes into a large active complex and causes the degradation of DNA in nuclei. Hence cleavage of DFF45 is an important step in apoptosis.
