Antibodies to Frizzled proteins 1-10

Introduction: Frizzled proteins are seven-transmembrane receptors which act as receptors for Wnt proteins.

Wnt proteins form a family of highly conserved secreted signalling molecules that regulate cell to cell interactions during embryogenesis. The N-terminal cysteine rich domain (CRD) region of Frizzled receptors is involved in the ligand-receptor interaction. The binding of a Wnt ligand to Frizzled (done with the aid of cofactors) activates Frizzled. This, in turn, activates a cytosolic protein called Dishvelled. Activated Dishvelled inhibits the ß-catenin degradation complex, so ß-catenein escapes by proteasomes and enters the nucleus where it binds to the promoters and / or enhancers of its target genes.

At present, at least ten different human Frizzled genes have been described, playing essential roles in the regulation of embryonic development, tissue and cell polarity, formation of neural synapses, and the regulation of proliferation. The signalling pathway has been shown to be implicated in the genesis of numerous human cancers.
