Antibodies to Protein Phosphatase 2

Introduction: Protein phosphorylation is dependent on the opposing actions of protein kinases and protein phosphatases. Four major protein phosphatases have been identified in the cytosol of eukaryotic cells which are responsible for the dephosphorylation of serine and threonine residues in proteins. Although all 4 protein phosphatases, PP1, PP2A, PP2B, and PP2C, have overlapping substrate specificities in vitro, they can be distinguished by the use of inhibitor proteins and by their dependence on metal ions.

Protein Phosphatase 2 is one of the four major Ser/Thr phosphatases, and it is implicated in the negative control of cell growth and division. It consists of a common heteromeric core enzyme, which is composed of a catalytic subunit and a constant regulatory subunit, that associates with a variety of regulatory subunits. The B regulatory subunit (PP2A, PP2CB, PPP2CB) might modulate substrate selectivity and catalytic activity.

Protein phosphatase 2C(PP2C alpha) is a Mn2+- or Mg2+-dependent protein serine/threonine phosphatase that inhibits the human stress-responsive p38 and JNK MAPK pathways and regulates cellular stress response in eukaryotes.

Samples: Whole cell lysate from HeLa (5, 15 and 50 mcg for WB; 1 mg for IP, 20% of IP loaded), 293T (T; 50 mcg), and mouse NIH3T3 (M; 50 mcg) cells. Antibodies: Affinity purified rabbit anti-PP2A antibody NB 100-2260 used for WB at 0.1 mcg/ml (A) and 1 mcg/ml (B) and used for IP at 3 mcg/mg lysate (B). Detection: Chemiluminescence with exposure times of 3 minutes (A) and 30 seconds (B).

Fig.3: Mouse liver cytosol extract was resolved by electrophoresis, transferred to PVDF membrane and probed with anti-PP2Calpha antibody SM6005 (1:250). Proteins were visualized using a goat anti-mouse secondary antibody conjugated to HRP and a DAP detection system. Arrow indicates PP2Calpha (~43kDa).
