Western Blot of HRP conjugated anti-Goat IgG antibody R1318HRP showing detection of 50ng of Goat IgG (Lane 1) but not Human IgG (Lane 2). Samples were separated by 4-20% SDS-PAGE under reducing conditions and transferred to nitrocellulose membrane. The blot was blocked overnight at 4°C in 5% BSA in TBS. A 1/5,000 dilution of antibody in Blocking Buffer for Fluorescent Western Blotting was used to probe the membrane at RT for 1 h. The image was developed using Chemiluminescent FemtoMax™ Super Sensitive HRP Substrate (p/n Femtomax-020) for one minute. Rabbit HRP

Mouse IgG (H+L chain)

Western Blot of Mouse IgG Antibody Peroxidase Conjugated detecting mouse IgG. Mouse IgG (1.0 mg) was separated on a 4-20% gradient gel and then transferred to Whatman Protran BA 85 nitrocellulose (0.45 µm pore size) and blocked with 5% BLOTTO (p/n B501-0500) o/n at 4°C. The membrane was then probed with a 1/2,500 dilution of HRP Rabbit anti-Mouse IgG [H&L] R1253HRP for 45 min at RT. FemtoMax Super Sensitive Chemiluminescent HRP Substrate Was added and the image was captured using a BioSpectrum Imaging System with a BioChemi 500 -28C cooled 4.0 Megapixel CCD Camera from UVP (www.uvp.com, Upland, CA). Binning: 5X5. Exposure time: 2 min. Aperture open. No emission or excitation filters used. Rabbit HRP

Western Blot showing detection of 0.1 µg of recombinant p27 protein. Lane 1: Molecular weight markers. Lane 2: MBPp27 fusion protein (arrow; expected MW: 73.3 kDa). Lane 3: MBP alone. Protein was run on a 4-20% gel, then transferred to 0.45 µm nitrocellulose. After blocking with 1% BSA-TTBS (MB-013, diluted to 1X) overnight at 4°C, primary antibody was used at 1/1000 at RT for 30 min. HRP-conjugated Goat-Anti-Rabbit R1454HRP secondary antibody was used at 1/40,000 in MB-070 blocking buffer and imaged on the VersaDoc™ MP 4000 imaging system (Bio-Rad). Goat HRP
