Western blot of rabbit anti-Beta-galactosidase (E. coli) antibody biotin conjugated. Lane 1: Beta-galactosidase. Load: 50 ng per lane. Primary antibody: Beta-galactosidase (E. coli) antibody biotin conjugated Cat.-No. R1064B) at 1:1,000 overnight at 4°C. Secondary antibody: HRP streptavidin secondary antibody at 1/40,000 for 30 min at RT. Blocking buffer ( for fluorescent Western blotting) for 30 min at RT. Beta-galactosidase tag antibody

Western blotting using  fluorescein conjugated anti-Beta-galactosidase  tag antibody shows a band at ~117 kDa (lanes 1-3) corresponding to 60 ng, 30 ng and 15 ng, respectively of b-Gal present in partially purified preparations (arrowhead). Lane 4 shows no cross reactivity with proteins present in a non-specific control E.coli lysate. Proteins were resolved on a 4-20% Tris-glycine gel by SDS-PAGE and transferred to nitrocellulose and blocking using blocking buffer for bluorescent Western blotting. The membrane was probed with Fluorescein conjugated anti-Beta-galactosidase Cat.-No. R1064F diluted to 1/10,000. Reaction occurred for 2 hours at RT. Molecular weight estimation was made by comparison to a prestained MW marker in lane M. Fluorescence image was captured using the VersaDoc® imaging system. Beta-galactosidase tag antibody

Western blotting using Beta-galactosidase antibody Cat.-No R1064PS. Lane 1 shows 80 ng and lane 2 shows 20 ng loaded onto gel. Results for N<em>on-Reducing Conditions</em> of SDS-PAGE prior to transfer to nitrocellulose are shown on the left side of the figure; results obtainined under <em>Reducing Conditions</em> are shown on the right. Blots were blocked overnight at 4°C with blocking buffer for fluorescent Western blotting (p/n MB-070). The membrane was probed with Beta-galactosidase antibody Cat.-No R1064P diluted to 1/10,000. Reaction occurred overnight at 4°C. Dylight649™ conjugated goat-anti-rabbit IgG was used for detection. Molecular weight estimation was made by comparison to a prestained MW marker (center). Western blot using Anti-Biotin Ligase EPITOPE TAG antibody shows detection of the BLT Ligase Target in lysates of whole cell Bacillus subtilis strains producing either BAP-tagged (Biotin-Acceptor Peptidetagged) SpoIIIAH (XWB49) (~ 25 kDa) or BAP-tagged SpoIIQ (XWB46) (~33 kDa).
