Alkaline phosphatase from E.coli
Application Suitable for Immunoblotting (Western or Dot blot), ELISA, Immunoperoxidase electron microscopy and Immunohistochemistry as well as other peroxidase-antibody based enzymatic assays.
Western blot: 1/500 - 1/2,000
ELISA: 1/4,000 - 1/20,000.
This product has been assayed against 1.0 µg of alkaline phosphatase [E.coli] in a standard capture ELISA using ABTS as a substrate for 30 minutes at room temperature. A working dilution of 1/3,500 to 1/15,000 of the reconstitution concentration is suggested.
Background Alkaline phosphatase (ALP, ALKP) is a hydrolase enzyme responsible for removing phosphate groups from many types of molecules, including nucleotides, proteins, and alkaloids. The process of removing the phosphate group is called dephosphorylation. All mammalian alkaline phosphatase isoenzymes except placental (PALP and SEAP) are inhibited by homoarginine, and, in similar manner, all except the intestinal and placental ones are blocked by levamisole. Heating for ~2 hours at 65°C inactivated most isoenzymes except Placental isoforms (PALP and SEAP).
Caution Do NOT add Sodium azide!
Concentration 1.0 mg/ml (by UV absorbance at 280 nm)
Storage Store lyophilized at 2-8°C for 6 months or at -20°C long term.
After reconstitution store the antibody undiluted at 2-8°C for one month or (in aliquots) at -20°C long term. Avoid repeated freezing and thawing.
Shelf life: one year from despatch.
Horseradish peroxidase
Multi-step process including delipidation, salt fractionation and ion exchange chromatography followed by extensive dialysis against the buffer
Buffer System:
0.02 M Potassium phosphate, 0.15 M Sodium chloride, pH 7.2
0.01% Gentamicin sulfate (Do NOT add Sodium azide!)
10 mg/ml BSA (immunoglobulin and protease free)
Lyophilized purified Ig fraction
Restore with 0.1 ml of deionized water (or equivalent).
This antibody detects alkaline phosphatase [E.coli]. No cross reactivity occurs against mammalian forms of alkaline phosphatase.
Assay by immunoelectrophoresis resulted in a single precipitin arc against anti-peroxidase, anti-rabbit serum as well as purified and partially purified alkaline phosphatase [E.coli].
