Application The antibody can be used for flow cytometry. Excitation 488 nm, peak emission 575 nm.This antibody has been titred for application in flow cytometry, 1 ?g is the maximum amount of reagent required for 1 x 106 cells. For studies on platelets care should be taken to avoid activation of platelets during drawing of blood. Platelet activation markers are weakly expressed on platelets prepared from directly fixed normal blood. The expression is max. on thrombin (1 U/ml) stimulated, washed platelets. Fix cells or platelets with 1% paraformaldehyde, wash and adjust to approx. 108/ml for further incubations. See for further instructions for use A., flow cytometry / FACS analysis.
Background CD62P is a member of the small selectin family of cellular adhesion molecules, which also includes CD62E and CD62L. Its structure, similar to the other members of the selectin family, consists of an N terminal lectin like domain of C type, followed by an epidermal growth factor like motif, a series of short consensus repeats, a transmembrane domain, and a cytoplasmic tail. The CD62P antigen is a 140 kDa glycoprotein, located in the alpha granules and the dense granules of platelets and endothelial cells. Activation of these cells results in rapid mobilization of CD62P from the storage granules to the cell surface. Activated platelets have a stable CD62P expression, while the endothelial cells lose CD62P expression within 1 h of activation, because of endocytosis of the molecule. CD62P is also expressed on megakaryocytes, but resting platelets and endothelial cells show no surface staining of CD62P.

The monoclonal antibody is directed against the CD62-antigen, (an integral mem?brane sialoglycoprotein 140 kD) also designated GMP-140 (granule membrane protein 140) or PADGEM, which is expressed in platelet alpha granules that are translocated to the platelet surface upon activation with strong agonists. A simular protein, present in endothelial cell Weibel-Palade bodies, is transiently expressed on the surface of activated endothelial cells and functions as adhesion molecule for granulocytes.
