Solubilized keratin extracted from human stratum corneum.
Application Immunohistochemistry on frozen and paraffin embedded sections: 1:40-1:50. This antibody can be used on formalin-fixed, paraffin embedded tissue sections. Prolonged fixation in buffered formalin can destroy the epitope. We recommend an incubation time of 30 minutes at 37C when used in conjunction with Bio/medas immunoperoxidase staining systems. For optimal results, use high temperature antigenic unmasking technique (see instructions below) or predigest the sections with Auto/Zyme (M34) before the application of the primary antibody. The antibody may be used at a dilution of 1:40 to 1:50 with AutoProbe III (Cat. No. 08-803). It is recommended that this product be used on frozen tissue sections or specimens. Western Blot: 1/50 - 1/200. Recommended positive control: Skin, Squamous Granulocytes Carcinomas. Other applications not tested. Optimal dilutions of this antibody are dependent on conditions and should be determined by the user.

Storage Store the antibody at 4°C. Do not freeze! Shelf life: one year from despatch.

Aliquoting Instructions: Do not dilute the entire reconstituted solution at once. Withdraw aliquots as needed with a micropipette and keep concentrated stock at 4C. Dilute according to the particular application being used. In general, the 0.05M Borate pH 8.0 containing 0.15M Sodium Chloride, 0.05% Sodium Azide, is a good dilutent to use with most antibodies. Avoid diluting the entire contents of the vial at once since the diluted solution may have reduced stability.
This antibody reacts with 66 kD and 57 kD polypeptides in extracts of stratum corneum. However, in two dimensional immunoblots, this antibody identifies keratins of molecular weights 68, 58, 56.5 and 50 kD, which corresponds to keratins No. 1, 5, 10 and 14 respectively according to Molls classification. Reacts with Human and Rabbit. Others not tested.
