Immunogen:
Synthetic Lipid A
Application Agglutination (use undiluted).
ELISA (1:10 in phosphate buffered saline containing 4% BSA and 0.05% Tween, see also protocol below).
Concentration ~0.1 mg/ml (OD280nm, E1%=14)
Protocols For ELISA prepare bacteria suspension in PBS (5x10e8 bact./ml).
Coat ELISA plate with 100 µl/well for 1 hour at 37°C, store overnight at 2-8°C and wash 5 times with tapwater with 0.05% Tween (TT).
Incubate with diluted antibody, 1 hour at 37°C.
Wash 5 times with TT.
Incubate with appropriate conjugate (anti-mouse Ig enzyme labeled antibody).
Add substrate solution, incubate, stop reaction and read optical density.

Reactive with the hydrophilic region of Biphospholipid A (LPS gram negative bacteria, Lipopolysaccharide) in PHA and ELISA. In PHA, clone 26-5 induced hemolysis only with SRBC coated with Lipid A and not with ReLPs.
