Background Peripherin is a class III intermediate filament subunit protein found in certain classes of neuron, most of which are located in the peripheral nervous system. The peripherin gene can be alterately spliced, producing three protein products known as Per-58, Per-56 and Per-61, the numbers reflecting the apparent molecular weight of each protein on SDS-PAGE analysis (1). The Per-58 form is the most abundant isotype. Per-56 has a truncated C-terminus when compared to the Per-58 form, while the Per-61 has a 32 amino acid insert within the a-helical coiled coil region of the molecule. The Per-61 form is unable to assemble into 10nm filaments and recent data suggests that it is in fact neurotoxic. Interestingly, peripherin protein is upregulated in the lower motor neurons that degenerate in Amyotrophic Lateral Sclerosis (ALS), focusing attention on peripherin as a potential contributory factor to this serious human neurodegenerative disease. Indeed, it has recently been suggested that specific overexpression of the the Per-61 isoform may be causative of some cases of human ALS.
