Background Tetracycline repressor protein class B from transposon Tn10 (TetR) is a transcription regulator that represses the tetracycline resistance element. The affinity for the tetracycline resistance gene (TetA) promoter operator sites are reduced by the binding of tetracycline to TetR. TetR also binds to DNA on its N-terminal region due to its helix-turn-helix structure (HTH tetR-type DNA-binding domain). The tetracycline resistance element repressed by TetR is responsible for the maintenance of genetic antibiotic resistance traits in strains of Escherichia coli (E.coli). These traits confer an advantage in selection to E. coli expressing this antibiotic resistance (PMID: 18039823).
