Add. information When diluting test samples; note the dilution factor for future data analysis. For example, if you diluted your sample 1:2 (250uL serum sample into 250uL sample diluent) and this yielded an OD of 0.437, then when read off the curve the OD of 0.437 correlated to 200 pg/mL, you would simply multiply 200pg/mL by 2 (your dilution factor) yielding a value of 400pg/mL. This indicates that the original starting sample had 400pg/mL of analyte present.

Background General Serum Diluent (SD1) is specifically formulated for the dilution of mammalian and chicken serum samples in antigen-down ELISA formats. Sample diluents are used to dilute serum or antigen samples into the functional range of the assay. Due to the finite binding capacity of the coated ELISA plate surface, highly concentrated samples may overload this capacity, thus requiring dilution prior to testing in the assay. Hyperimmunized serum samples contain nonspecific antibody that may interfere with the titration. Sample diluents can also reduce background noise associated with nonspecific bridging of signal-generating conjugates to the plate well surface. Proprietary additives have been incorporated into SD1 that minimize nonspecific binding interactions of the specific IgG in the samples, thus decreasing background noise. SD1 also contains a buffered BSA protein base to assure a constant pH and solute environment favorable to antibody-antigen interactions. - Can be used for antigen-down ELISAs to detect antigen-specific immunoglobin in the sera of mammals and chickens. - May be used to dilute an antigen standard and samples used in sandwich ELISA formats. - Contains additives to reduce nonspecific binding interactions. - Contains an antimicrobial agent to provide a solution free of bacterial contamination. - Formulated at 1X, ready to use. - pH 7.4 at 1X, ionic strength.
