Antibodies to SET1, SET7 and SET7/9 Histone Methyltransferase Family - FocusOn 040

Introduction: Eukaryotic DNA is assembled with histone proteins to form nucleosomes, which are the basic repeating unit of chromatin. The methylation of lysine residues of histones plays a critical role in the regulation of chromatin structure and gene expression. Acetylation, phosphorylation and methylation of the amino-terminal tails of histone are thought to be involved in the regulation of chromatin structure and function. The SET family of Methyltransferases play a critical role in Methylation of Histones.

SET1A and SET1B are Histone methyltransferase that specifically methylates 'Lys-4' of histone H3, when part of the SET1 histone methyltransferase (HMT) complex, but not if the neighboring 'Lys-9' residue is already methylated. H3 'Lys-4' methylation represents a specific tag for epigenetic transcriptional activation. The non-overalpping localization with SETD1A suggests that SETD1A and SETD1B make non-redundant contributions to the epigenetic control of chromatin structure and gene expression. They specifically tri-methylates 'Lys-4' of histone H3 in vitro.

SET2 is a Histone methyltransferase that methylates 'Lys-36' of histone H3. H3 'Lys-36' methylation represents a specific tag for epigenetic transcriptional activation. It probably plays a role in chromatin structure modulation during elongation via its interaction with hyperphosphorylated POLR2A and binds DNA at promoters. SET2 may also act as a transcription activator that binds to promoters and binds to the promoters of adenovirus 12 E1A gene in case of infection, possibly leading to regulate its expression.

SET7/9 is a Histone methyltransferase that specifically monomethylates 'Lys-4' of histone H3. H3 'Lys-4' methylation represents a specific tag for epigenetic transcriptional activation. It plays a central role in the transcriptional activation of genes such as collagenase or insulin and is recruited by IPF1/PDX-1 to the insulin promoter, leading to activate transcription. SET7/9 has also methyltransferase activity toward non-histone proteins such as p53/TP53, TAF10, and possibly TAF7 by recognizing and binding the [KR]-[STA]-K in substrate proteins. It monomethylates 'Lys-189' of TAF10, leading to increase the affinity of TAF10 for RNA polymerase II, and 'Lys-372' of p53/TP53, stabilizing p53/TP53 and increasing p53/TP53-mediated transcriptional activation.

SM6006 Immunoblot analysis: Recombinant SET7/9 was resolved by electrophoresis, transferred to PVDF membrane and probed with anti-SET7/9 (1/1000). Proteins were visualized using a goat anti-mouse secondary antibody conjugated to HRP and a DAP detection system. Arrow indicates SET7/9 (~50 kDa).
