Marker for Axonal Growth Cones - FocusOn 012

Introduction

Growth cones are specialised sensory structures located at the tip of neurites, which are involved in the mechanism that enables elongation and navigation of axons during neuronal development. Growth cone motility depends on cytoskeletal dynamics. The two major cytoskeletal components in growth cones are actin filaments, which are predominantly located in the peripheral (P)-domain, and microtubules in the central (C)-domain. Spatially localized actin polymerization and depolymerization and actin-myosin interactions generate retrograde movement of actin filaments, which is viewed as a force-generating system to pull the growth cone forward. Growth cones express various cell adhesion molecules (CAMs) that recognize localized guidance cues present on neighboring cells or in the extracellular matrix and translate them into a directed axonal extension. One important axonal CAM is L1, which belongs to the Ig superfamily. L1 serves as both a ligand and a receptor. Therefore, L1-NCAM/CD171 antibodies may serve as a good marker to stain axonal growth cones.
