Background of CD298 / ATP1B3 antibody

Na + /K + -ATPase is an integral membrane protein complex that is responsible for the ATP-dependent transport of sodium and potassium across the cell membrane. Na + /K + -ATPase hydrolyzes ATP to maintain the transmembrane gradients of Na + and K + . The Na + /K + -ATPase consists of a large transmembrane catalytic subunit, termed the a subunit, and an associated smaller glycoprotein, termed the ß subunit. There are four isoforms of the a subunit ( a 1, a 2, a 3 and a 4) and three isoforms of the ß subunit ( ß 1, ß 2 and ß 3) in mammalian cells. All a subunits are 10-membrane-spanning proteins that contain the binding sites for Na + , K + and ATP. The ß subunits are single-membrane-spanning glycoproteins that are required for the proper folding and trafficking of the Na + /K + -ATPase from intracellular stores to the plasma membrane. The ß subunit is essential for the normal activity of the enzyme, and it appears to be involved in the occlusion of K + , and the modulation of K + and Na + affinity of the enzyme. Different combinations of the a and ß subunits can alter the kinetics of enzyme activity. Substantial differences in the tissue and cell specificity of expression have been found for each a and ß subunit.
