Background of Claspin antibody
Claspin is a cell cycle regulated adaptor protein which peaks at the S/G2 phase and plays an essential role during checkpoint-mediated cell cycle arrest, in response to DNA damage or inhibition of DNA replication.
Following replication stress, the phosphorylation of threonine 916 and serine residues 945/982 on claspin, enables its association with Chk1 (cell cycle checkpoint kinasomologue 1), a protein kinase responsible for the initiation of G2 arrest in response to DNA damage. Claspin has also been shown to interact with ATR (the ATM- and Rad3-related protein kinase) and Rad9, further checkpoint proteins involved in Chk1 activation.
