Background of Claudin-7 / CLDN7 antibody
The Claudin superfamily consists of many structurally related proteins in humans (1). These proteins are important structural and functional components of tight junctions in paracellular transport (1,2). Claudins are located in both epithelial and endothelial cells in all tight junction-bearing tissues (1). Three classes of proteins are known to localize to tight junctions, including the Claudins, Occludin and Junction adhesion molecule (3). Claudins, which consist of four transmembrane domains and two extracellular loops make up tight junction strands (4). Claudin expression is highly restricted to specfic regions of different tissues and may have an important role in transcellular transport through tight junctions (5). mRNA studies indicate that Claudin-7 is specifically expressed in mouse lung and kidney, but not in heart, brain, spleen, liver, skeletal muscle or testis (6). The gene encoding human Claudin-7 maps to chromosome 17p13 (7).
Immunohistochemistry (IHC) analyzes of Claudin7 antibody (Cat.-No.: AP06067PU-N) in paraffin-embedded human lung carcinoma tissue.
Human HeLa; WB Suggested Anti-CLDN7 Antibody Titration: 0.2-1 ug/ml. ELISA Titer: 1:312500. Positive Control: Hela cell lysate; CLDN7 antibody - C-terminal region (ARP31357_P050) in Human HeLa cells using Western Blot
Human Lung; WB Suggested Anti-CLDN7 Antibody Titration: 0.2-1 ug/ml. ELISA Titer: 1:312500. Positive Control: Human Lung; CLDN7 antibody - middle region (ARP33613_P050) in Human Lung cells using Western Blot
CLDN7 antibody (C-term) (Cat. #AP10180b) immunohistochemistry analysis in formalin fixed and paraffin embedded human prostate carcinoma followed by peroxidase conjugation of the secondary antibody and DAB staining. This data demonstrates the use of the CLDN7 antibody (C-term) for immunohistochemistry. Clinical relevance has not been evaluated.
CLDN7 Antibody (C-term) (Cat. #AP50951PU-N) flow cytometric analysis of Hela cells (right histogram) compared to a negative control cell (left histogram).FITC-conjugated goat-anti-rabbit secondary antibodies were used for the analysis.
Immunohistochemical analysis of Claudin 7 staining in human lung cancer formalin fixed paraffin embedded tissue section. The section was pre-treated using heat mediated antigen retrieval with sodium citrate buffer (pH 6.0). The section was then incubated with the antibody at room temperature and detected using an HRP conjugated compact polymer system. DAB was used as the chromogen. The section was then counterstained with haematoxylin and mounted with DPX.
