Background of DIABLO / SMAC antibody

Apoptosis is a conserved cell suicide program essential for the development and homeostasis of multicellular organisms. Abnormal inhibition of apoptosis is a hallmark of cancer and autoimmune diseases, whereas excessive cell death is found in neurodegenerative disorders such as Alzheimers disease. Executioners of the apoptotic program are cysteine proteases termed caspases that exist as inactive zymogens in living cells and are activated during apoptosis. Active caspases cleave key intracellular protein substrates, resulting in the characteristic morphological changes associated with apoptosis. The release of cytochrome c from the mitochondria triggers the oligomerization of Apaf-1 in an ATP/dATP-dependent manner and induces the autoactivation of caspase-9. Active caspase-9 in turn activates downstream effector caspases including caspase -3, -6 and -7.
Given the potentially irreversible cascade triggered by the activation of initiator caspases, it is crucial that activation of caspases be tightly regulated. A number of cellular proteins have been shown to modulate caspase activation and activity. These include members of the Inhibitor of the Apoptosis Protein (IAP) family, such as XIAP, c-IAP-1 and c-IAP-2, which block cell death by inhibiting the activity of the effector caspases -3 and -7 and the initiator caspase-9. Smac/DIABLO (Second Mitochondria-derived Activator of Caspase) is a mitochondrial protein of 27 kDa that is released from mitochondria during apoptosis. Smac/DIABLO promotes caspase activation through binding and neutralizing IAPs.
