Background of M-CSF / CSF-1 antibody

M-CSF (macrophage colony stimulating factor), also known as colony stimulating factor-1 (CSF-1), is a homodimeric glycoprotein the subunits of which are linked by disulfide bonds. Different splice variants of M-CSF have been found which differ at the C-terminus. M-CSF is the primary regulator of the survival, proliferation and differentiation of monocytes and macrophages. M-CSF synergises with IL-1, IL-3 and IL-6 in the stimulation of proliferation and the differentiation of primitive haematopoietic cells into macrophages. In normal human macrophages M-CSF induces antibody-dependent cellular cytotoxicity, enhances counts of monocytes, leads to an expansion of neutrophilic granulocytes and supports osteoclast differentiation. In both monocytes and macrophages, M-CSF induces the synthesis of IL-1, G-CSF, IFN, TNF, plasminogen activator, thromboplastin, prostaglandins and thromboxanes as well as oxidative metabolism. This is a HCX protein. HCX Expression System Details HCX proteins mimic the proteins in the human body because they are expressed from human, rather than animal, insect or bacterial cells. This process gives them human post-translational modifications. Recombinant DNA techniques allow a human protein with the correct amino acid sequence to be expressed in a non-human cell line. However, non-human cells lack the appropriate cellular machinery, such as specific glycosyltransferases, necessary to produce the correct human post-translational modifications of a protein. An extreme example is seen in E. coli cells, which produce recombinant proteins with no glycosylation, as the above figure illustrates. Rodent and yeast cells are able to glycosylate proteins, but they are still different from glycosylation in human cells.
