Background of NFAT2 / NFATC antibody

The nuclear factor of activated T-cells (NFAT) transcription complex is required for the expression of a group of proteins that collectively coordinate the immune response. Two forms of NFAT, the cytosolic (NFATc1) and the pre-existing NFATc2 (formerly NFATp), have been recognized on separate genes, and show sequence similarity with the Dorsal/Rel family of transcription factors. Studies indicate that NFATc is required for interleukin (IL-2) gene expression, is found primarily in lymphoid tissues and is induced upon T-cell activation. NFATc2 shows 73% homology with NFATc1 within the Rel-similarity region, but has a wider tissue distribution and does not appear to be inducible. T-cell activation requires two signals for initiation, one from the antigen receptor and the other by accessory cells or agents which activate protein kinase C. Upon binding of the antigen to the T-cell receptor, several membrane events are initiated which are similar to those initiated by many hormones, including the mobilization of intracellular calcium stores. NFATc1 appears to be independently modified by agents which activate protein kinase C or increase intracellular calcium, indicating one function of NFATc1 might be to integrate signals derived from different cell membrane receptors. Studies indicate that NFATc1 is also regulated by calcineurin, a calcium and calmodulin dependent protein phosphatase. Dephosphorylation is essential for NFATc activation, nuclear translocation, and early gene expression in T-cells. The immunosupressive drugs, cyclosporin A and FK506 when bound to the immunophilins cyclophilin and FKBP, respectively, inhibit dephosphorylation and activation of NFATc1 by calcineurin.
