Background of NPY receptor 1 / NPY1R antibody

Neuropeptide Y Receptor Y1 (NPY1R) is a G-Protein Coupled Receptor containing characteristic 7 transmembrane spanning domains of the Neuropeptide Y (NPY) receptor family that includes NPY2R and NPY5R; it is located at human chromosome 4q31.3-32 (1). It is activated by the binding of NPY, a neurotransmitter in the central and peripheral nervous system (2). Activation of NPY1R is generally associated with reduction of cAMP accumulation, increase of intracellular free calcium concentration and modulation of MAPK pathway via several signaling molecules, including the protein kinase C (PKC) (3). The presence of NPY1R is important in the migration of endothelial cells and its activation also stimulates cell proliferation (3, 4). NPY1R is therefore a potential new molecular target in cancer treatment.
NPY is suggested to play an important role in tumor biology based on the high density of NPY receptors in many cancers (5). Among the five NPY receptors (NPY1R-5R), NPY1R is highly expressed in and its activation is important in the proliferation of cells in cortical tumors, prostrate cancer, Ewing sarcoma family of tumors, synovial sarcomas and on blood vessels in many other sarcomas (5, 6). NPY1R is also expressed in heart, colon, kidney, brain, adrenal gland and placenta. NPY1R is approximately a 56kDa protein (384 amino acids), the unexpected larger size of the mature protein is probably due to post-translational modifications including several glycosylation sites near the N-terminal end of the protein.
