Background of Placenta growth factor / PGF antibody

PlGF (Placental growth Factor) was isolated initially as a cDNA from a human placenta cDNA library. PlGF is expressed also in human umbilical vein endothelial cells colon and mammary carcinomas. Placenta growth factor (PlGF) is a member of the vascular endothelial growth factor (VEGF) family of growth factors. PlGF and VEGF share primary structural as well as limited amino acid sequence homology with the A and B chains of PDGF. The biologically active form of this protein is a disulfide-linked dimer. The N-glycosylated dimeric protein is secreted and stimulates the proliferation of endothelial cell lines and supports angiogenesis. PlGF has been renamed PlGF-1 after the discovery of PlGF-2, which has a 21 amino acid insertion not present in PlGF. PlGF-1 and PlGF-2 compete in a dose-dependent way with the 165 amino acid form of VEGF for receptor binding on endothelial cells. The PlGF proteins bind with high affinity to Flt1, but not to Flk1/KDR. Neuropilin-1 probably functions as a receptor for PlGF-2. This is a HCX protein. HCX Expression System Details HCX proteins mimic the proteins in the human body because they are expressed from human, rather than animal, insect or bacterial cells. This process gives them human post-translational modifications. Recombinant DNA techniques allow a human protein with the correct amino acid sequence to be expressed in a non-human cell line. However, non-human cells lack the appropriate cellular machinery, such as specific glycosyltransferases, necessary to produce the correct human post-translational modifications of a protein. An extreme example is seen in E. coli cells, which produce recombinant proteins with no glycosylation, as the above figure illustrates. Rodent and yeast cells are able to glycosylate proteins, but they are still different from glycosylation in human cells.
