Background of PRDM9 / PFM6 antibody

PR domain containing 9 protein (PRDM9) is one of at least 20 PR domain containing molecules in human (1). The PR domain is a derivative of SET domain which has been known to act as catalytic domain for histone lysine methylation (2). PRDM9 is located on human chromosome 5p14, it contains a PR domain in its N-terminal portion and a C2H2 type zinc finger motif in its C-terminal portion (3). A PR domain’s catalytic activity for histone methylation is dependent upon the individual proteins of that PR domain. The catalytic activity of PRDM9 towards histone H3 is restricted only for tri-methylation of lysine 4 in histone H3 (H3K4) from di-methylation (4). Tri-methylation of H3K4 is a characteristic trait of transcriptionally active genes (5); therefore, PRDM9 is often implicated in transcriptional regulation. PRDM9 also plays an essential role on proper progression of meiotic prophase in mouse germ cell lineage (4).
Recent study has reported that PRDM9 is expressed in early meiosis and it controls the activation of mammalian recombination hotspots (6). Mammalian meiotic recombination occurs at these hotspots, ensuring the orderly segregation of meiotic chromosome and thus creating genetic variation among offspring. Therefore, deficiency of PRDM9 in mice resulted in sterility in both sexes (6). PRDM9 mutations in human can cause failure in spermatogenesis and thus idiopathic infertility in human males (7). PRDM9 is expressed in germ cells entering meiotic prophase. It is approximately a 46.1kDa protein (384 amino acids).
