Background of U2AF2 / U2AF65 antibody

The study of pre-mRNA splicing and of spliceosome assembly was benefited from the biochemistry in mammalian systems, the combination of facile genetics and biochemistry available from yeast Saccharomyces cerevisiae, and by comparing the two systems. There are clear differences in the splice site recognition in mammals and yeast systems; these include some of the earliest interactions required for intron-exon recognition and the involvement of SR proteins, enhancer elements and U2AF proteins. Splicing factors in S.pombe compared ot S. cerevisiae, typically are more similar to their mammalian counterparts, e.g. the snRNAs and many U2AF proteins (1). Furthermore, S.pombe contains SR proteins and highly conserved homologs of both subunits of U2AF, which are critical for early interactions in the mammalian system. The splicing components recognize sequence elements in the pre-mRNA through an ordered series of interactions. The first of these form the early or commitment complex (E or CC) and are ATP independent. In this complex, the 5′ splice site (SS) is recognized by U1 snRNP.
The Schzosacharomyces pombe splicing factor U2 auxiliary factor (U2AF59) is comprised up of 2 interacting subunits of 59 and 23kDa which is similar to human homologue of 65 and 35 kDa. The fission yeast cDNA library screening using yeast two-hybrid system for the interaction of proteins with U2AF59 identified several partners including hsU2AF35 subunit. U2AF59 is a pre mRNA splicing factor. The SF1-U2AF59-U2AF23 complex has a role in the recognition of the branch site (5’-UACUAAC-3’), the pyrimidine tract and the 3; splice site at the 3’ end of introns. U2AF59 forms homo dimmer and heterodimer with U2AF small subunit (U2AF23), the small subunit U2AF23 and SF1(bpb1) interact to form a complex required for complex A formation that interact with pop3. U2AF59 is a nuclear protein and belongs to the splicing factor SR family which contains 2 RRM (RNA recognition motifs). The difference between human and yeast U2AF proteins is the lack of lacks an arginine/serine-rich region in the yeast protein. The interaction motifs of these proteins are same in both human and yeast (2).
